Phagocyte energy metabolism as target to induce remyelination in multiple sclerosis. (Research)
Abstract
Phagocytes are key players in the pathology of multiple sclerosis (MS). Their faulty reprogramming upon engulfing excessive myelin disables repair processes in the brain. Rewiring energy metabolism is highly regulated in myelin-clearing phagocytes and provides novel targets to halt MS progression. In this project, changes in energy metabolism will be studied in myelin-loaded macrophages in vitro and in chronic active MS lesions. Next, the therapeutic potential on skewing macrophages towards a reparative phenotype by targeting metabolic enzymes will be evaluated in preclinical models of MS.
